Structural Analysis of Protein-Protein Interactions in the Polyadenylation-Complex by Chemical Crosslinking and Mass Spectrometry

Messenger RNA precursors (pre-mRNAs) undergo different processing steps, including splicing, 5’-end capping and 3’-end cleavage, and polyadenylation. Pre-mRNAs get cleaved downstream of the poly(A) signal (often AAUAAA) and polyadenylated (up to 250 adenosin residues) by a multi-protein complex, which consists of different subcomplexes. The complete compositon, structure and mechanism of the polyadenylation machinery is still unclear. To adress this issue, the subcomplexes are analysed by chemical cross-linking in combination with high resolution with mass spectrometry.
